Gut Microbiota Regulate Motor Deficits and Neuroinflammation in a Model of Parkinson’s Disease

چکیده
The intestinal microbiota influence neurodevelopment, modulate behavior, and contribute to neurological disorders. However, a functional link between gut bacteria and neurodegenerative diseases remains unexplored. Synucleinopathies are characterized by aggregation of the protein α-synuclein (αSyn), often resulting in motor dysfunction as exemplified by Parkinson's disease (PD). Using mice that overexpress αSyn, we report herein that gut microbiota are required for motor deficits, microglia activation, and αSyn pathology. Antibiotic treatment ameliorates, while microbial re-colonization promotes, pathophysiology in adult animals, suggesting that postnatal signaling between the gut and the brain modulates disease. Indeed, oral administration of specific microbial metabolites to germ-free mice promotes neuroinflammation and motor symptoms. Remarkably, colonization of αSyn-overexpressing mice with microbiota from PD-affected patients enhances physical impairments compared to microbiota transplants from healthy human donors. These findings reveal that gut bacteria regulate movement disorders in mice and suggest that alterations in the human microbiome represent a risk factor for PD.

Ellagic Acid Protects the Brain Against 6-Hydroxydopamine Induced Neuroinflammation in a Rat Model of Parkinson’s Disease
Introduction: Neuroinflammation may play as an important risk factor in progressive degeneration of dopaminergic cells. Antioxidants have protective effects against free radicalsinduced neural damage in Parkinson’s disease (PD). In the present study, we examined the effects of ellagic acid (EA) on locomotion and neuroinflammatory biomarkers in a rat model of PD induced by 6-hydroxidopamin...
متن کاملGut Microbiota Dysfunction as Reliable Non-invasive Early Diagnostic Biomarkers in the Pathophysiology of Parkinson’s Disease: A Critical Review
Recent investigations suggest that gut microbiota affects the brain activity through the microbiota-gut-brain axis under both physiological and pathological disease conditions like Parkinson's disease. Further dopamine synthesis in the brain is induced by dopamine producing enzymes that are controlled by gut microbiota via the microbiota-gut-brain axis. Also alpha synuclein deposition and the a...
متن کاملThe effect of nobiletin on performance of rats in forced swimming and elevated plus maze tests in intranigral lipopolysaccharide rat model of Parkinson's disease
Background and Objective: Anti-inflammatory property of nobiletin (NOB) is proven and neuroinflammation is involved in triggering and progression of neurodegenerative disorder such as Parkinson's disease (PD). PD is a neurodegenerative disorder characterized by motor and non-motor features including psychiatric symptoms such as depression and anxiety. The purpose of this study to investigate wh...
متن کاملChanges in the composition and function of the gut microbiota in celiac disease
Evidence is supported the hypothesis that any changes in the composition and function of the gut microbiota play a fundamental role in a number of chronic inflammatory diseases including celiac disease (CD). In the last decade, several culture-independent methods have been developed to identify the components of the human microbiome. The study of microbiota based on nucleic acid analysis found ...
متن کاملExternal human exposure and management immune system in pathogenesis of irritable bowel syndrome
External exposed radiation may play an important role in pathogens of irritable bowel syndrome (IBS), although is thought to arise due to a combination of genetic and environmental factors. The result is dysregulated immune responses due to alteration in the gut microbiota population and the subsequent development of gut inflammation. It has recently been shown that the effect of ioni...
